05. Immunohistochemical diagnosis of porcine epidemic diarrhea (PED)

There was a sudden outbreak of epidemic diarrhea of piglets in Japan, with principal features of watery diarrhea, dehydration and high mortality in newborn animals. This disease could not be distinguished clinically, pathologically and electron microscopically from transmissible gastroenteritis(TGE). The piglets were not diagnosed based on the virology of TGE. Immunohistochemically, specific antigens of the porcine epidemic diarrhea(PED) virus were detected in the cytoplasm of enterocytes by streptavidin-biotin(SAB). Coronavirus particles were confirmed in the intestinal contents of the piglets by transmission electron microscopy, but the PED virus could not be isolated. The SAB is thus shown of potential use for the diagnosis of PED.
