20. Modification of Oligosaccharide of Bovine Interferon-tau in the Baculovirus Expression System

Japanese

Recombinant bovine interferon-tau (rboIFN-t) bearing manmarian-type N-glycan are expressed by the baculovirus expression system. rboIFN-t were expressed with Sf beta 4Gal/alpha 2,6ST cells (stably transformed cell line that expressed beta-1,4-galactosyltransferase and alpha 2,6-sialyltransferase). rboIFN-t strongly reacted with ConA, RCA-120 and SNA in lectin bloting. It indicates that N-glycan of rboIFN-t contains mannose, galactose and sialic acid. In this result, "mammalianized" foreign glycoprotein with N-glycan containing galactose and sialic acid was produced in the baculovirus expression system.
